科研成果

已对160余种药用植物进行了深入系统的化学和生物活性研究,发现了大量的新化合物和新骨架类型分子,以及超过200多个具有重要生物活性的分子(代表性分子见图1);阐明了相关药用植物的化学成分和部分药用植物的药效物质基础;总结归纳和提出了一批新骨架分子的生源合成路线,并通过化学合成和转化对部分生源路线进行了验证;将化学合成修饰和生物活性研究相结合,对系列重要活性分子进行结构优化和构效关系研究,发现了系列药物先导和多个候选药物;完成了系列新颖结构和重要活性分子的全合成研究。部分代表工作如下:

1逆转肿瘤多药耐药脂环肽类物质的发现:脂环肽是一类罕见的天然化合物,具有独特的结构和广泛的生物活性,长期深受天然产物、有机合成和生物合成领域的广泛关注。从楝科药用植物海南樫木中发现了一个新骨架1-元环脂肽类化合物(图2),其结构确定是一挑战性的工作。同时,该分子能显著逆转癌细胞的多药耐药,针对多个耐药细胞株,其倍数逆转率在28至1040范围;进一步的作用机制研究表明该化合物是通过抑制P-糖蛋白(P-gp)的转运功能而克服多药耐药。该研究发表在JACS(2019, 14, 6812-6816)。

2、二萜维生素缀合物的发现与合成:从香港樫木中发现了一类天然二萜维生素C缀合物(命名为hongkonoids A-D,图3),是国际 发现的萜类维生素C缀合物,结构中包含一个独特的5/5/5-三环螺缩酮丁内酯单元以及一个二萜长链。该类分子生源上可能是从维生素C牻牛儿基牻牛儿基焦磷酸缩合物,经过Claisen重排作为关键步骤,以及系列的修饰转化而产生。受此启发,我们通过一个会聚式的策略,以5.4%至9.6%总产率,12至14步反应,完成了对hongkonoids A-D全合成。其关键步骤是成功利用一锅法的Claisen重排、脱保护以及5-exo-trig串联环化反构筑了该类化合物的母核。生物活性测试结果显示,该类化合物及其合成衍生物中的部分化合物对NF-κB、11?-HSD1和甾醇合成显示出活性不等的抑制作用,其中活性更好 的一个化合物在两种巨噬细胞RAW 246.7和BMDM中对LPS诱导的炎症反应均有较显著的抑制活性。该研究发表在JACS(2018, 140, 2485-2492)。

3、强效免疫抑制新骨架三萜化合物的发现:从大戟科植物海南叶下珠中发现了一类新骨架三萜化合物(phainanoids A–F),在小鼠淋巴细胞的非特异性毒性作用及增殖实验中,发现这类化合物有很强的抑制小鼠淋巴细胞增殖的作用,且部分化合物的活性比临床上使用的环孢甲素更强,如化合物phainanoid F (图4),其抑制T淋巴细胞增殖作用是环孢甲素的7倍,抑制B淋巴细胞增殖作用是环孢甲素的221倍。是迄今为止,从药用植物中发现免疫抑制活性最强的一类化合物。同时,研究还对这类分子的构效关系进行了初步探讨。该研究发表在JACS(2015, 137, 138-141)。
